Comprehensive mapping of long-range interactions reveals folding principles of the human genome.

چکیده
We describe Hi-C, a method that probes the three-dimensional architecture of whole genomes by coupling proximity-based ligation with massively parallel sequencing. We constructed spatial proximity maps of the human genome with Hi-C at a resolution of 1 megabase. These maps confirm the presence of chromosome territories and the spatial proximity of small, gene-rich chromosomes. We identified an additional level of genome organization that is characterized by the spatial segregation of open and closed chromatin to form two genome-wide compartments. At the megabase scale, the chromatin conformation is consistent with a fractal globule, a knot-free, polymer conformation that enables maximally dense packing while preserving the ability to easily fold and unfold any genomic locus. The fractal globule is distinct from the more commonly used globular equilibrium model. Our results demonstrate the power of Hi-C to map the dynamic conformations of whole genomes.

متن کاملA Simple Genome Walking Strategy to Isolate Unknown Genomic Regions Using Long Primer and RAPD Primer
Background: Genome walking is a DNA-cloning methodology that is used to isolate unknown genomic regions adjacent to known sequences. However, the existing genome-walking methods have their own limitations. Objectives: Our aim was to provide a simple and efficient genome-walking technology. Material and Methods: In this paper, we dev...
متن کاملStatistical Analysis of Long-Range Interactions in Proteins
We carry out a comprehensive study of long-range interactions on a large data set of non-homologous proteins. Our study reveals that the long-range interactions between amino acids far apart are common in protein folding, and play an important role on the formation of secondary structure. Using residue-wise contact order(RWCO) to describe long-range interactions, we further evaluate the effect ...

Centralizing ethics in sustainability has led to an evolutionary process towards ethical approach as a holistic approach of sustainable development. This approach, deep in concept, has evolved sustainability and endures the most fundamental relations of human and environment. The most recent definition of ethical sustainability, describes this approach as a complex of human relations, including...
متن کاملCharacterization of hundreds of regulatory landscapes in developing limbs reveals two regimes of chromatin folding.
Complex regulatory landscapes control the pleiotropic transcriptional activities of developmental genes. For most genes, the number, location, and dynamics of their associated regulatory elements are unknown. In this work, we characterized the three-dimensional chromatin microarchitecture and regulatory landscape of 446 limb-associated gene loci in mouse using Capture-C, ChIP-seq, and RNA-seq i...
